Any character can be major, just as any character can be sidelined. If we were holding this conversation in the 1990's then "major" characters would include Sleepwaker, Deathlok and Darkhawk. If we held this conversation in 2013 then Star-Lord, Rocket, Groot, Drax and Gamora are absolute D list. One year later and they are known throughout the world and are now A list. In the context of modern Marvel - Negasonic Teenage Warhead is as relevant as Tigra, Wonder Man, Annihilus or any other character who had their time in the spotlight.
Here is a list of characters/groups who were created to be completely throwaway or just to serve a corporate purpose but to whom most people will think they were created to be a major IP asset of Marvel Comics:
The Avengers - Martin Goodman saw the success of the Justice League of America and ordered Stan Lee to replicate it. Stan (and more importantly Jack Kirby) created the Fantastic Four in response. To Goodman's amazement - the FF was a hit. He went back to Stan Lee and said "do that again!" - once Marvel had created yet more succesful titles Stan Lee was able to finally carry out his original brief and "copy the Justice League". The first issue of the Avengers contains no original material/characters at all, even the villain is recycled. The Avengers are an empty, artifical construct copied from a rival. But I do love them!
Alpha Flight - created to fight the X-Men, never intended to be used again. They were designed with zero backstory and John Byrne had to be cajoled into bringing them back as a standalone title. The initial issue $30,000 payment probably helped!
Deadpool - Rob Liefeld threw Deadpool at Bob Harras along with a bunch of other designs with an offer to "Use 'em, trash 'em whatever, give me a call!" Deadpool is a rip-off of Deathstroke the Terminator but also according to Liefeld his "mutant version of Spider-Man".
Wolverine - designed to be a one-use foe for The Hulk. Was used in X-Men solely because he was Canadian in a new X-Men relaunch. Wolverine would have been killed off early in Chris Claremont's run if John Byrne had not become artist and wanted to "save the Canadian"
Mar-Vell/Carol Danvers/Ms Marvel/Captain Marvel - Mar-Vell was created so that Marvel Comics could steal the trademark of "Captain Marvel" from DC Comics. Carol was created as a protection of the tradesmark " Captain Marvel". Ditto for She-Hulk - she was created to prevent the Hulk TV show from introducing a female Hulk and then not only owning the copyright but also devaluing The Hulk trademark. Ditto Spider-Woman.
Sure, there are certain characters who transcend trends like Spider-Man and Superman and who will always probably be popular but at the same time the fan base once got tired of both the X-Men and the Hulk to the point both were cancelled.

I am not saying that all 4*s are bad. Just that additional time/money spent on making (for devs) or collecting (for players) new 4*s offers a vanishingly small return. New 4*s are either useless because theybare redundant in a tier of 100+ (like this character, or misty, or anti-venom) or sufficiently powerful as to render all other 4*s useless. That's a bad deal for players either way.
