This matters when you're low on roster space. From my guide, If you run out of roster space, consider saving up your standard/heroic tokens. If you open up a new character with a full roster, you won't have the space to use it, so you can stash them until you have space. If you're cover starved though (in that you have your characters at max level and need more covers), you should probably just bite the bullet and use up the tokens.0

Nobody knows for sure, but the general thought is that there's probably not code that changes the probability based on how many you open at once. That is, you have the same chance of getting a character you want by opening them one at a time, as you do saving them up and opening a whole bunch at once.
Some people like saving them up so that in any one session of opening tokens you're more likely to find one that makes you happy (because you're opening so many). Other people like opening them one at a time for instant gratification. That's pretty much just a question of personal preference.
As far as I can see, there are three meaningful ways you can optimize:
- if there's a tournament going on right now and there's a cover you know is in a token that would help you do significantly better (for example, getting 3* Thor right now would be great if you don't have him at all, since it will help you in both the pvp tournament and the main event), you should open all your tokens right away.
- if there's a limited time event token that has a higher concentration of characters you need (for example, the highly trained tokens have psylocke, black panther, and 3* thor, who are all quite recent and so most players need all three), as compared to the heroic token, you should open it right away. If it has a lower concentration of characters you need than the heroic token (for example, the old Widowmaker special event token only had black widows in it, so if you had all three black widows at max covers), you should wait until it expires and turns into a regular heroic token.
- if your roster is developed to the point where you have most or all of your covers maxed, you should wait until new characters get added, so that each token has a higher chance of giving you someone that you need. This one is a bit tricky; if you're in this situation you usually want to open tokens as soon as a character you really want gets added, unless the next new character is also someone you really want. For example, I held all my tokens to wait for Psylocke, and then waited for Black Panther, and then waited for 3* Thor. Now that they're all in and the next character is Captain America, who I care less about, I opened all the tokens I was hoarding.
This is all assuming that you have enough roster space to keep any new heroes that you get! If that's not the case, then you should follow Polarity's advice above.
