Save your isos for Punisher.
If you get more than a cover for Punisher isos will need to make it competitive.

You've gotta level that 4* Wolverine dude. You know how awesome the 4*s are.

Rags, Spidey, and Mags of the ones you have. Punisher looks like he could be very good too.

I've taken to keeping a 10k buffer at all times -- I'll level up various heroes as I'm using them, but that lets me get some new buffed character into something vaguely useful quickly, or splurge on a character I want to level up after getting a surprise cover.

Edit: Of course, even if you win all 3 Punisher covers his max level will be in the 20s at best unless you drop HP on improving his powers too.
